Hempen was unable to coach Sunday's match because he received two yellow cards in Friday's 1-0 loss at Missouri. But he still received credit for Sunday's win.
Hempen became one of 17 active coaches in Division I soccer to reach the 250-win plateau. He is in his 21st season overall and eighth at CU.
Hempen wasn't available Sunday for comment.
The Buffs improved to 9-2-2 overall and 3-1-1 in the Big 12 in beating Kansas (8-4-1, 2-2).
Colorado senior Nikki Keller scored CU's first goal, with the assist coming from Nikki Marshall.
"It wasn't the prettiest game we've ever played, but the effort was there," Keller said. "We were able to pull off two goals in the second half to win it."
The Buffs began the two-game road trip the tough loss at Missouri.
"We bounced back from the game on Friday," Keller said. "We were definitely disappointed, but winning the game 2-0 today gave us our confidence back, so we'll be ready to go next weekend."
Colorado's Gianna DeSaverio gave the Buffs some breathing room just under 10 minutes after Keller's goal with her sixth goal of the season for a 2-0 lead.
Colorado goalkeeper Kirstin Radlinski shutout the Jayhawks for the first time this season.
Next up for CU, the Buffs host No. 12 Texas at 4 p.m. Friday at Prentup Field.
